Is there a way to lock layers? I was hoping I could animate things inside a filled shape or an alpha lock.

Animation inside an alpha mask is not possible on the iPad. An animation inside a shape would work if you use the eraser on a layer that covers the entire workspace with the same background color.
Here is an example with black layer on a black background.

There are A LOT of fun things you can create using background colors and erasing parts. You can do the eraser method on the fly in TT, or you can create very complex png files using identical background colors.
